m6A 甲基 RNA 定量試劑

在真核細胞中，N6-methyladenosine (m6A) 是 RNA 修飾作用中存在比例最多的一種，m6A 修飾作用經由 methyltransferase complex METTL3 反應而成，另外去除甲基的機制則是由 m6A RNA demethylases 家族的 FTO 及 ALKBH5 兩種蛋白來進行，過程中需要α-ketoglutarate (α-KG)- 和 Fe2+ 等分子介入。目前實驗發現 METTL3、FTO及ALKBH5 在生物體中扮演很重要的角色，從發育、代謝到生殖都和這些蛋白息息相關，這三種蛋白引起的 m6A 修飾作用，在整體 RNA甲基修飾的種類中佔了大於80%，主要發生在 mRNA 中，mRNA 的 m6A 會影響 RNA 代謝過程，例如 splicing、nuclear export、轉譯能力及穩定度等，若 m6A RNA methylase 和 demethylase 如果發生缺失，有可能造成 RNA 機能不正常以及疾病的產生。對於此種動態且可逆性的 RNA m6A 甲基修飾作用，或許可作為疾病的新一代 epigenetic marker。此產品的實驗原理是先將 total RNA 利用 RNA high binding solution 結合至 strip well 中，而後使用 N6-methyladenosine antibody 及 detection antibody 來測定 N6-methyladenosine (m6A)，detection antibody 經實驗試劑反應後會在 450 nm 波長處產生訊號，此時即可使用光譜儀進行測定定量，試劑組中包含 postitive control 及 negative control。
